EZ-Cholesterol Assay Kit 는 free cholesterol과 cholesteryl esters 각각 또는 모두를 흡광 (colorimetric) 및 형광 (fluorometric) 측정을 통한 정량 분석법을 제공합니다. 혈액의 대부분의 cholesterol은 cholesteryl esters의 형태로 존재하는데 이는 cholesterol esterase에 의해 cholesterol로 가수분해 됩니다. Cholesterol은 다시 cholesterol oxidase에 의해 산화되면서 부산물로 H2O2를 생성하고 이는 Cholesterol probe와 반응하여 발색 (OD570 nm) 및 형광 (Ex/Em = 535/595 nm)을 일으킵니다. cholesterol esterase가 존재 하는 반응에서는 total cholesterol (cholesterol 그리고 cholesteryl esters)을 분석하며 cholesterol esterase가 존재하지 않는 반응을 통해서는 free cholesterol을 분석할 수 있습니다. Cholesteryl ester는 total cholesterol 분석 값과 free cholesterol 값의 차로 구합니다.
